I am thinking no, just because in 3.02 (The Kids Are Alright) Sam mentions that the YED had killed anybody that Mary even knew, like Relatives, Friends, ect. But they could talk more about Sam and Dean family members that haven't been mentioned before, such as Mary and John's Relatives.
Still, it is just a theory. ;p
